Paroxysmal Nocturnal Hemoglobinuria (PNH) – CD55/59 Erythrocytes, FLAER Granulocytes/Monocytes

Specimen Requirements:

  • 2 x 5 - 10 mL peripheral blood in a lavender-top tube (EDTA)
  • Peripheral blood only; it must be received in the lab within 24 of collection hours, but no later than 72 hours.

Specimen Storage/Transport Temperature: The fresh peripheral blood specimen can be transported on cold pack or wet ice, but do not fix or freeze specimens (no dry ice).

Unacceptable Conditions
  • Specimens greater than 24 hours old
Stability (collection to initiation of testing):
  • Ambient: 24 hours
  • Refrigerated: 24 hours
  • Frozen: Unacceptable

Use:

This test is used to diagnose PNH and quantify PNH clones in blood. PNH may be associated with hemolytic anemia, leukopenia, thrombocytopenia and aplastic anemia. This method uses expression of CD55/59 on erythrocytes and the more sensitive fluorescent aerolysin (FLAER) on granulocytes/monocytes.
